Abnormal phosphoenolpyruvate transport in erythrocytes of hereditary spherocytosis.

چکیده
The rate of phosphoenolpyruvate transport in erythrocytes from patients with hereditary spherocytosis and from healthy individuals was examined in a 0.1 M citrate buffer (pH 6.1 at 37 degrees C) containing 10 mM phosphoenolpyruvate and 10 mM NaF. The rate in erythrocytes from patients with hereditary spherocytosis was 0.09 +/- 0.02 mumol/min/ml of cells (mean +/- SD, n = 8), whereas the rate in erythrocytes from healthy individuals was 0.23 +/k- 0.03 mumol/min/ml of cells (mean +/- SD, n = 8). The lower rate of transport in erythrocytes of hereditary spherocytosis could not be described to a different glycolytic response to NaF or to a difference in cell age distribution. This phenomenon seemed to be a reflection of some abnormality in the erythrocyte membrane from patients with hereditary spherocytosis.

By A. WILLIAM SHAFER T HE BASIS for the abnormal morphology of the erythrocytes in hereditary spherocytosis ( HS ) has intrigued investigators for many years. Prankerd and Altman’ and Prankerd, Altman and Young2 reported that after a relatively long period of incubation ( 1-6 hours ) with p32 orthophosphate, the rate of laI)eliflg of adenosine triphosphate (ATP) and 2, 3-diphosphoglycerate ( DP...

The incorporation of extracellular orthophosphate-32P into cellular ATP, 2,3-diphosphoglyceric acid, and inorganic phosphate has been measured over a period of 6 hours in vitro in red blood cells from normal subjects and from patients with hereditary spherocytosis who had undergone splenectomy. The pattern of labeling of the intracellular compounds was found to be the same in both types of red ...

Background: Neonates affected by hereditary spherocytosis may suffer from significant jaundice. This study was conducted on neonates with jaundice hospitalized at the Children’s Hospital in Bandar Abbas, South Iran, to determine the frequency of hereditary spherocytosis among them. Patients and Methods: In this cross-sectional study, 814 neonates with jaundice hospitalized at the Children’s ...
متن کاملComparative Study of the Amount of Re-released Hemoglobin from α-Thalassemia and Hereditary Spherocytosis Erythrocytes
Hemoglobin release test (HRT), which is established by our lab, is a new experiment to observe the re-released hemoglobin (Hb) from erythrocytes. In this study, onedimension HRT, double dimension HRT, and isotonic and hypotonic HRT were performed to observe the re-released Hb from the blood samples of normal adult, hereditary spherocytosis (HS), and α-thalassemia. The results showed that compar...

Studies during the last three decades have enabled the development of detailed molecular insights into the structural basis of altered function in various inherited red cell membrane disorders. This review highlights our current understanding of molecular and mechanistic insights into various inherited red cell membrane disorders involving either altered membrane structural organization (heredi...
